Isolation of specific bacteriophages - Pseudomonas savastanoi pv. glycinea - and their use in soybean bacterial blight control

Bacterial blight is one of most harmful diseases of legumes, reducing the profitability of soybean production in Russian Federation. Among a number of Pseudomonas isolates obtained from diseased seeds and plants of soybean, 4 strains were selected and confirmed as Pseudomonas savas-tanoi pv. glycinea (Psg). Properties of the isolated bacteria were similar to type strain of Psg CFBP 2214 in plant virulence, LOPAT tests, and PCR analysis for coronafacate ligase gene, and partly -in the phage reaction profile.

During the phytosanitary inspection of a lot of stored potatoes of cv. Zhukovskiy ranniy in the Orel region of Russia, tubers showed signs of lesions in the form of local darkening of the flesh without apparent softening and fungal structures on the surface (coremia). A pure culture of the pathogen was isolated and the morphological characteristics of the conidial sporulation structures in the extracted isolate were described; the average sizes of vegetative mycelium, conidia, conidiophores and coremia were given.

This article describes the application of the classical PCR method for the diagnosis of the soybean bacterial blight pathogen Pseudomonas savastanoi pv. glycinea in seeds. Two methods for pathogen isolation from infected soybean seeds, five methods for DNA extraction and two master mixes for preparation of reaction mixture were investigated. The use of a PCR-based test system with oligonucleotides specific for the coronaphacate ligase (cfl) gene allowed the diagnosis of the soybean bacterial blight pathogen in seeds at a concentration of 2 × 103 CFU/ml.

The paper presents the results of the analysis of diseases of infectious fallout of winter wheat at the Field Experimental Station of the Russian State Agrarian university – Moscow Timiryazev Agricultural Academy from 2013–2015 to 2020–2021, depending on the tillage technology and weather conditions. It was shown that the main disease was pink snow mold (caused by Microdochium nivale). The analysis of the dynamics of the disease over 5 years showed that the prevalence was higher under zero tillage than under moldboard tillage (6.6 and 3.9%, respectively).

Evaluation of the antibacterial properties of essential oils and plant extracts against potato blackleg pathogens

The antibacterial activity of 25 samples of essential oils and seven aqueous samples and ethanolic plant extracts was evaluated against the complex of potato blackleg pathogens Dickeya chrysanthemi, Pectobacterium odoriferum, Dickeya didantii and Pectobacterium carotovorum subsp. carotovorum. To identify the described species, studies of pathogens were carried out in vitro with a skin diffusion test on agar, requiring minimal inhibitory (MIC) and minimal bactericidal concentrations (MBC) of essential oils and plant extracts.
