The San Jose scale, Diaspidiotus (Quadraspidiotus) perniciosus (Comstock, 1881), is a significant economic pest of fruit crops regulated under the Common List of Quarantine Pests of the Eurasian Economic Union. In quarantine phytosanitary laboratories, diagnosis of this species is typically performed using morphological methods; PCR teats for D. perniciosus are not routinely employed. This article describes the development of PCR tests for the identification of D.

Among more than 20 existing types of longspine sandbur, (Cenhrus longispinus (Hackel ex Kneuck.) Fernald), an annual cereal of North American origin, is of particular importance for the Russian Federation. This plant is widely distributed in temperate countries as a weed plant. It contaminates farm crops, vegetables and melons, vineyards and gardens. It is a quarantine object for the Russian Federation and the countries of the Eurasian Economic Union. On the territory of our country there are small disparate foci localized along railway lines and within urban areas.

Optimisation of molecular-genetic methods for diagnosing fungi of Genus Sclerotinia

Phytopathogenic fungi are the causative agent for disruptive crop plant diseases, besides causing significant losses in crop yield annually. Sclerotinia is one of the most common and dangerous pathogens. Species of the Sclerotinia genus cause widespread disease across a broad range 40 of economically important agricultural crops. Regular screening of Sclerotinia in plant material is necessary due to wide host-plants range and absence of resistance to this phytopathogen.

Detection of the soybean bacterial blight pathogen Pseudomonas savastanoi pv. Glycinea in seeds by the PCR method

This article describes the application of the classical PCR method for the diagnosis of the soybean bacterial blight pathogen Pseudomonas savastanoi pv. glycinea in seeds. Two methods for pathogen isolation from infected soybean seeds, five methods for DNA extraction and two master mixes for preparation of reaction mixture were investigated. The use of a PCR-based test system with oligonucleotides specific for the coronaphacate ligase (cfl) gene allowed the diagnosis of the soybean bacterial blight pathogen in seeds at a concentration of 2 × 103 CFU/ml.
